Pig's brain function restored hours after death

Pig's brain function restored hours after death

Using a synthetic blood solution and pump which simulates the heart, researchers at America's prestigious Yale University showed that brain cells presumed dead began showing signs of life, long after they were presumed unrecoverable.

The pig brains came from the pork industry; the animals were not raised in a lab for this experiment, yet Yale scientists were so concerned the pigs might become conscious that they gave drugs to the disembodied brains to reduce any higher brain activity.

The surprise findings challenge the idea that cellular death within the brain is a swift and irreversible process.

In the new study, Sestan's team wanted to find out what happens in the brain after blood flow stops and test whether they could restore function to a dead brain in a way similar to the brain cells in petri dish. First, the specially formulated solution - the "artificial blood" - that the team used does not contain the same makeup found in human blood.

It's the first time neuroscience researchers have seen an effect to this degree but Vissel noted the study also may not be entirely surprising to neurologists - they have been aware for years that areas of the brain blocked off from blood supply by clots during stroke can be recovered if the clot is removed within four hours. All of this carried on until 10 hours after the decapitation.

"What if we could, in a future iteration of this method, introduce - without cutting the head off, but just by introducing catheters - perfuse the brain earlier on in the resuscitative process", he said.

It also reconfirms the notion that consciousness dissipates quickly within minutes of brain death and the only way to stop that from happening is to restore circulation in the brain immediately.

"Secondly, could brains maintained by BrainEx help scientists discover new treatments for brain diseases like Alzheimer's and Parkinson's disease, which are proving really hard to treat?"

The pigs' brains, however, did not ever regain consciousness.

Researchers hope their BrainEx system - detailed Wednesday in the journal Nature - can help scientists study cellular functionality inside intact large mammalian brains.

"This is not a living brain, but a cellularly active brain", Sestan said in a press conference.

However, the researchers, who study brain development and evolution, observed that the small tissue samples they worked with routinely showed signs of cellular viability, even when the tissue was harvested multiple hours postmortem.

There was no electrical activity across the brain which is the benchmark for brain death, let alone the coordinated firing of the billions of neurons required for consciousness. We found no evidence that these brains have any activity which is associated with perception or consciousness.

The team will now work on applying the research to patients who have had a stroke and those who have been patients declared brain dead. The study, backed by the National Institutes of Health, offers no immediate clinical breakthrough for humans, according to the authors.

The technique, known as BrainEx, has fuelled debate over whether definitions of brain death will have to be reconsidered.

"Cell death in the brain occurs across a longer time window than we previously thought", Sestan said.

"An important fact, since restoration of blood flow to the rest of the body (following a cardiac arrest, for instance), may activate injury processes in non-brain tissues which produce substances that damage the brain (for example, through activation of inflammation)". "For most of human history, death was very simple", Christof Koch, president and chief scientist of the Allen Institute for Brain Science in Seattle, said in a Nature article accompanying publication of the Yale study.

Scientists urged people not to let the study put them off organ donation. There may need to be "new rules", she said.
